This project involves functional characterization of CALR mutations identified in the patient samples. The study will use FLAG tagged Calr protein to identify the sub-cellular localization of mutant Calreticulin. In addition using Co-IP technique it is planned to search for binding partners of mutant Calreticulin relative to WT Calreticulin. As Calreticulin is known to act as Calcium buffer in the endoplasmic reticulum, analysis of Calcium signalling changes with mutant calreticulin will be undertaken. This study will shed light on novel role of Calreticulin in haematological malignancies like AML.
